For maturation of bovine oocytes, the following defined medium was used: MEM-α supplemented with follicle stimulating hormone, 1 μg/ml, human chorionic gonadotropin, 0.3 U/ml, Na-pyruvate, 0.2 mM. Using this protein-free culture system complemented with hormones, there were received 23.1% of parthenogenetic, 15.6% reconstructed and 15.7% control embryos (fertilized in vitro) on blastocyste stage. It has been shown that a method of activation of oocytes (natural with use of spermatozoons, or artificial, by Са++ ionophore А23187), and methodical details of reconstruction of cells did not affect percent of the embryos which have been developed to preimplantation stages. The data obtained suggest that serum protein is not a necessary component of medium for high-grade nuclear and cytoplasmic in vitro maturation of bovine oocytes with intact cumulus. The tested protein free medium allows to receive a cytoplasts of stable quality for their use in animal cloning research.
